Feature request: gene-level liability score for knockdown/knockout #4550

Open

Description

@AR-Shicheng

Hi @opentargets team — thanks for building and maintaining Open Targets' safety and genetics tooling (gentropy included); it's been a genuinely useful reference for how to structure on-target liability data, and it's saved us a lot of design time.

Would you consider a small feature for target liability evaluation? The idea: not every gene is safe to inhibit or knock down. A lightweight workflow could take a gene name as input and return (1) a human safety score for knockout/knockdown, and (2) the supporting safety signals behind it — essentiality, genetic constraint, critical-organ expression, on-target effects seen in genetics or drug data. Where red flags show up, that's a signal the gene may only tolerate partial inhibition or need tissue-restricted delivery rather than full knockdown.

Would appreciate it if the team could consider adding this — thanks for thinking it over.

    Hi @AR-Shicheng , target safety is an important issue and we are also working on this.
    Could you please provide a bit more detail on how exactly it is evaluated and what should be used as input?
    Depending on the input, Gentropy may not be the best place for this - if it uses many data sources (such as expression, animal models, etc.) it may be better placed in the platform ETL rather than in Gentropy.
